The transcriptomic profiling of ACE2 across 13 tissues (119 cell types) reveals that ACE2 expresses in lung AT2, liver cholangiocyte, colon colonocytes, esophagus keratinocytes, ileum ECs, rectum ECs, stomach epithelial cells, and kidney proximal tubules. The low expression of ACE2 in the lung raises the possibility of existence of co-receptors facilitating nCoV infection.

The single-cell co-expression profiling of 451 genes across the 13 human tissues (119 cell types) reveals that 94 genes are co-expressed with ACE2. Of those, ANPEP, ENPEP, and DPP4 are the top three genes correlated with ACE2. The result suggests that CoVs infect similar types of cells and CoV-infected patients share similar clinical symptoms.

"CellPhoneDB" analysis revealed that the nCoV-targeted cells (ACE2-expressing) interact with several cell types in different tissues, especially macrophages in the lung, liver, and stomach, through CD74-MIF interaction and other signaling pathways. Other than MIF gene, PROs, SCs and TAs showed high interactions in the ACE2-expressing cells in ileum and rectum.

Respiratory tract scRNA-seq data analysis showed that pulmonary AT2 cells and respiratory epithelial cells had high ACE2 expression. The nasal and bronchial samples showed no ACE2 expression. Thus, the lungs are considered a high-risk organ for SARS-CoV-2 infections. Lungs were identified as the most vulnerable organ for SARS-CoV-2 infection, according to ACE2 expression. Urinary system scRNA-seq data analysis showed high ACE2 expression in Proximal tubule cells of kidney and bladder urothelial cells. The cluster of Kidney cells with high ACE2 expression also expressed canonical markers of kidney PT cells CUBN and LRP2.
